Anybody on the LOCI trial?

I’ve not come across any threads for this so thought I would start one as a place for us to share our journey.

I’m going in for an ultrasound and blood test tomorrow. Hopefully all will be ok and I’ll be accepted on the trial 🤞

I am on cycle 3 CD 6 just finished my course of three tablets of clomifene or letrozole but surprisingly had a spontaneous period this time which has not happened in a very long time hoping this may be the time I ovulate x

Hi ladies

So I tested yesterday after having some brown discharge (that’s now gone)… and I got a very faint first positive on cheapies and digital tests 🥹

I feel so anxious though because I took another one this morning and I feel like the lines are even fainter and the progression is not good enough? Everywhere I read it says hcg should double every 48 hrs and I just can’t stop thinking that I’m heading towards a chemical. I also know that I’m at least 12 DPO because of confirmed ovulation.

Going in for betas tomorrow but so so so anxious. What do you all think?

@Cherryblooms thank you. Had my app with the research nurses yesterday to give back the remaining meds and diary and answer some questions. They did advise that when I have follow up with consultant at end of this month they will unblind me and possibly allow us to explore the other ovulation med as you mentioned. We’re awaiting on a repeat test for partner to see if morphology has improved 🤞

@Cherryblooms lovely news. Congrats.

I know Iv finished the trial so just watching from afar but I had my follow up with consultant over the phone this morning and was unblinded. So thought I would share… so I was on clomifine and placebo. I thought I was on placebo as didn’t experience any stomach issues. I ovulated each cycle on lowest dose. Partner is having a repeat semen analysis as his morphology was 1% so hopefully has improved. Next steps for us she advised was if morphology has improved then option of another 3 months of ovulation induction (possibly either and metformin) or privately funded IVF or IUI. Lot to think about 🙃

Hi Everyone,

I just wanted to come here and share the sad news that unfortunately mine was a chemical pregnancy :( I went in for a confirmed blood test but hcg was very low (below 70) and never doubled. I have started some slight bleeding today as expected. I guess we are back to square one..

One thing that made me really upset when speaking to the team was that they said that I could not get unblinded until the whole trial was over (which could be a year from now). This goes against the information I have been verbally given before and this frustrates me as I feel like if I now know the dose that finally made me ovulate + pregnant, If I don't know if it was clomid/letro I have to start from the beginning to test again? Can anyone advise what to do here because from what I can see there has been several people in this thread that was unblinded after the 6 cycles were over?

I am so frustrated and sad this week as you can imagine. It just feels like bad news on top of bad news. The DR has not given me a call after the miscarriage either as they promised me. Ugh, the team is lovely normally but today I am really really tired.

@Cherryblooms im so sorry to hear your sad news. It’s really difficult but you need give yourself time to process it and go through all of the feelings.
I had a miscarriage just before the trial and it took a little while for my body to regulate before I could start the trial.

I’m sorry @Cherryblooms to hear your news. I’m not sure re the unblinding I was advised once the trial ended after 6 months I would be unblinded or if coming of trial early. Sending you lots of love
