Anybody on the LOCI trial?

I’ve not come across any threads for this so thought I would start one as a place for us to share our journey.

I’m going in for an ultrasound and blood test tomorrow. Hopefully all will be ok and I’ll be accepted on the trial 🤞

Not the best way to start my Monday.. expected my scan to be an improvement from the last but nope! I have a large cyst so no more scans until my next cycle.

Really trying to listen to my own words of trust the process but it's so difficult. I feel my body has let me down yet again.

😔 I’m sorry your news wasn’t what you hoped for either. Thinking of you 💗

It’s so frustrating and the endless merry-go-round can leave you feeling desolate.
I’m feeling okay now, but I certainly wasn’t when I posted last week.

Take some time for you, I had a lovely day on Friday, I spent the whole day with friends just having lunch, shopping and managing to shelf the infertility and enjoy myself.
I’ve had long conversations, sometimes they help sometimes they don’t.
I find comfort in knowing that one day, irrespective of the means, I will be a Mother; the treatment doesn’t guarantee the outcome we’d like but for me adoption will, be that from the UK or another country.
In an ideal world this would work, but the end goal of raising a child I will be blessed with and I hold onto that, in the darkest moments.

Good evening ladies,

My period was delayed this time (around 2 weeks) so I had a scan to see where we were at and then my period came on the 4th.
I started a double dose and had another follicle track on day 7 which showed vast improvement from the first attempt.
I had one 10mm follicle on the left and 2 on the right which were already at 17 and 18mm, which were suspiciously large for the time frame, so they wonder if those two are actually cysts. The womb lining was good too, so fingers crossed this does the trick.
I went on holiday so my next scan is the 21st, I hope this one is also going to show progress.
So for now I am still on the trial.
I had no side effects this time with the double dose at all, so I’m grateful for that.

Anyone else have an update? 🙏 praying for good news for you all x

Hiya! That's great news, really chuffed for you! How are you feeling about it?

I still haven't started the trial, my blood tests from last estimated ovulation show that I may have ovulated. So that want me to go for another scan for when they predict I will next ovulate.

I really don't know why they keep making me go, I have literally been having scans every month since January and only ovulated once. It's really starting to piss me off now.

😞@Rljnz It’s so frustrating, I know. Have they upped your dosage? How long do you have left of the trial?
I hope your next scan is more positive.

I had the scan and my lining is now at 4mm which isn’t ideal but they think I did ovulate as one of the follicles has ruptured so they estimate I ovulated on Sunday, their going to blood test on Monday to confirm ovulation.
It’s a rollercoaster because I’m aware that even if I did finally ovulate the lining may not be able to support implantation at only 4mm.
Just have to take each day as it comes.
Obviously they don’t know what I’m actually taking but they’d hazard a guess that I’m on the clomiphene and placebo judging by the thinning lining and no side effects.
They’ll review again after the bloods come back, as typically ladies who aren’t on the trail in my position would now be either given something to thicken the lining or changed to another drug which doesn’t have the thinning side effect - but the trail prevents them from being able to do this.

Hi ladies,

I had to take some time out since my last message. Finding out I had the cyst and another month of not knowing if I ovulate was tough. At 4.5 weeks I had strong pains in my stomach, they thought the cyst had twists or ruptured but I got scanned and my cyst was gone and I had period pains. I could of cried as this is my first natural period since I started the trial, my cycle lasted 35days rather than 3/4months.

I went for a scan on day 10, small follicle showing. I was there again today day 13, and the lining is slightly thicker and 1 follicle slightly bigger. They expect I will ovulate later in my cycle so I asked for another scan. I pray and hope they confirm I am ovulating on Monday. They do plan to up my dose to 3 tablets on my next cycle which should regulate things more.

@PCOSlife I hope you received some positive news at your scan

@Rljnz It's crazy how we are all on the same journey but the drugs are impacting us all differently. I never had any ovulation testing before the trial, I just had to wait 6 weeks since my last period before I could start provera then the trial drugs x

Hi everyone, i was part of the trial and ovulated straightaway but lost my baby at 16 weeks preterm labour just after stopping the medicines. I asked the trial nurse and she said it happens a lot. I got pcos.

Hi guys! Just been given my prescription and got all my tablets but I’m completely lost on what your supposed to take and when :( any tips? I’ve been given norethisterone or whatever it’s called to bring on a period so starting that tomorrow but unsure when to take the other tablets and how long for 😫

I went for another scan but my follicle hasn't grown much so they won't be counting this month. Next cycle is will be on 3 of the tablets and hopefully this will regulate ovulation!

@Tashyx that's exciting! Good luck and trust the process. I've been on the tablets since March and only now we think we're on the right meds. I would check with your fertility nurse but I was told, day 1 is when your properly bleeding and day 2 is when you will start your medication. 1 tablet for 5 days of the clomid/letrozole and Metformin is 1 tablet for a week, 2 tablets for the second week and 3 tablets after that x

@Tashyx my knowledge is the same as @Tryingforbabyb has written: red tablets from day 2, so you start your period then begin the tablets the next day for 5 days.
Then white tablets start alongside, every day but increase from 1 in the first week to 2 the second week and finally 3 thereafter and just keep taking them indefinitely, until hopefully baby is on board finishing at 14 weeks of pregnancy.

I had to read it a few times before I started to take them! It’s very daunting but it gets easier once you’ve had one go, which I hope is all you need 🩷

I’m on day 25 now, had my blood results today and confirmed ovulation.
If my period is on time it should be due on the 4th, but it was delayed last time so who knows. I’m also under a lot of stress right now as sadly my poor Dad is end of life care, so I’m deep in all that entails and it will be a miracle if my period isn’t affected by the stress. The two week wait is almost over for me, and I’ve not really had time to think too much about testing, which if my period doesn’t appear next week I’ll have to think about.
I have no tests, after the disappointment of last month I never replenished my stock.

I had no side effects taking the tablets this time, first go they were minimal and nothing after I finished taking them but this time I am having consistent breast soreness which has been there since my predicted ovulation date (around 11 days ago.)
My heads all over the place 😔 luckily I’m signed off work, so I can focus on my Dad but it does break my heart 💔

I’ll report back when I can x

I got a BFP!! 🥰 I still can’t believe it 🥲
I took two early response tests on Sunday.

The fertility clinic is going to scan me on the 25th of July.

Sending lots of love and prayers that everyone is also blessed with good news. 🙏🙏😘

Although they do advise against telling people so early I wanted to tell my Dad 🥹 so we ended up telling both sets of Grandparents, and our siblings so we’re are all over the moon!

I had headaches on my first round too, they stopped as soon I finished the red tablets x

My period is late by 4 days, have head aches, cold and flu like symptoms and a couple of bouts of diarrhea as well as being exhausted. BUT I am absolutely petrified of doings a pregnancy test. I don't think I can cope with a negative again 😔

Hi all.. I’ve been following this post for around 4 months now and I am finally ready to start the trial.

I have all of the meds and I am just waiting to start my period so I can take the tables on day two. I’ve taken provera to induce this, but it’s It’s been three days since completing the course and still nothing! I just want to start it now as it’s making be so anxious!
