Anybody on the LOCI trial?

I’ve not come across any threads for this so thought I would start one as a place for us to share our journey.

I’m going in for an ultrasound and blood test tomorrow. Hopefully all will be ok and I’ll be accepted on the trial 🤞

Can I ask what support you get for tracking ovulation , Scans ? Or just home testing as I have been advised to do?

@Pom99 I go to see the fertility nurses for a scan, usually day 10 and again at day 14.
I was told to stop the ovulation home tests because I was getting too fixated on them and there not accurate. I was to leave it to the nurses at my scan.

The only thing I track now is my bb temperature every morning but I'm not convinced how accurate this is either.

@Tryingforbabyb thank you for letting me know.
Seems as though everything is different in regards to what they get.
I unfortunately do not get any tracking scans, they told me i could fund these myself but the results wouldn't be taken into consideration as part if the trail.
i am using Clearblue and also the tiny little LH strips to test. Rather confusing as the results differ but managed to get a peak day CD 19 which they recorded.

@Pom99 that is so strange, even if I wasn't on this trial and I got Metformin I was still getting the tracking scans. As that's the only way they can confirm if I'm ovulating. Seems a good thing for me as I'm 5 months in but no record of ovulation yet so my 6month trial hasn't started yet.

I found premom app and ovulation tests reasonably priced but I've stopped them now x

Hi,

I’ve been a bit nervous about posting, but this thread seems really supportive. I’m on cycle three now, responded to the meds and confirmed ovulation first cycle. Which is of course great news, but the side effects just make me feel rubbish and my cycle is quite long so we end up having an almost three week wait.

I’ve not used any of the home ovulation kits and I’m unsure about it but, I feel a bit low about it all currently so have thought about it and was hoping for your guys thoughts?

Hi @EmJayRud your not alone! It is a rollercoaster.
Maybe try home tracking to see if you like it?
Someone on here recommended the Femom app to me and I am enjoying using this. I also track with the clear blue ( results different at first which confused me but then the app helped me understand some more.

Thank you, @Pom99 I did feel a bit more at ease once reading this thread.
I do use the period tracker app which I get on with but I’ve never done the home ovulation tracking as I never ovulated before the meds. Is the app specific to clear blue testing?

@EmJayRud I can only comment on what ive been doing so I am sure someone else in this thread will comment with tips that could help us both.
ive been using the clear blue advanced digital- this is what the nurse on the loci trail told me to use and is what they will record.
However I was advised my many people in a FB group called ‘Trying to conceive UK’ that they are known for incorrect readings so also to use the little LH testing strips .
so i did.
The CB gave me a static smile on day 16 which says peak but the same day i was showing 0% on the LH test strip (same pee sample) so i was so confused.
anyway LH strip cycle showed surge just out of no where in CD 19 or 20.
Glad i used both.

@Pom99 Thank you, I’ve had my tracking scans and blood work so I know I now ovulate but definitely don’t know what day, I have equally heard they can be unreliable.
I think we’ll give this third month a go and then maybe try the LH strips after, try and keep stress down. Thanks for much for your help 💕

Hey everyone!

So glad I searched this, been ttc for 5 years, sadly 4 miscarriages. Only managed to conceive once I had my endometriosis removed, now I can’t get past the 6 week mark.

Finally this year I got to see the fertility specialist, who said my ovary scan show pcos, but I have regular periods. She suggested I tried the loci trial.

im on my first month, being on 3 metformin/placebo. I had day 21 bloods done and the results came back at 37 which she said was high so I did ovulate. So fingers crossed this helps, I’m just worried that I may be able to catch but not get passed that 6 week mark. (Didn’t get told about scans so I’m not sure I’m entitled to it)

has anyone been through similar but managed to get to 12 weeks +?

sorry for the long winded message, it’s nice to be able to write something knowing others are going through the same thing!

Hi everyone! I’ve just started the trial and on my first cycle. Had my first scan on Friday (day 11) and had a dominant follicle of 17mm which I was happy about. Im using the Mira device and not ovulated yet but oestrogen is rising so I’m hoping it will be soon. Worried I won’t ovulate and need a trigger shot- the dr that scanned me suggested I have one but then realised I’m on the trial and said I couldn’t. Have people experienced having a dominant follicle on scan and then not ovulating? I should probably relax but worried I’m missing out on the trigger shot! Best of luck to you all xx

I think the worry is normal as we’ve all definitely been concerned about something at some point. I had good results from my first scan and then they gave me a day 21 blood test to confirm the ovulation, is this available to you? I was really worried, but the blood test put me at ease.

@EmJayRud @Louxxxx welcome to this crazy journey!
I've been on the tablets since March with no confirmation of ovulation. In July I took 3 Metformin/letrozole, at my day 10 and 14 day scan they said I was due to ovulate and went for bloods on day 21.
Obviously we tried had this cycle and it's paid off!!! We got a positive on day 27, I could cry, it doesn't feel real!!

@MyPCOSandme sorry I never got around to replying; my pregnancy hasn’t been the easiest, I have felt so unwell.
My Dad has sadly passed now, so this little blessing is a source of great comfort and hope for everyone.

I had the early 7 week scan, which was a really surreal moment and I have the 12 week scan in 9 days.

I’m pleased your pregnancy has not been as difficult, and hope the third trimester is going smoothly 🩷
They told me at my last assisted fertility appointment, before they handed me over, that we will be informed of the drugs we were given once the trial ends and the data is collected 😁 approx 2 years from now.

third trimester is going well. 58 days to go and I still don’t believe I am pregnant sometimes.

thanks for the update about when we will get the results. Once I got pregnant I never heard from the LOCI train team again. I can’t wait to receive the results and am happy to have taken part in this trail.

@EmJayRud thank you! It's easy for me to say now but I was extremely disheartened every cycle where they couldn't confirm ovulation etc. it will happen for us all!

@Tryingforbabyb everything we went through during the trail is all worth it in the end.

yes pregnancy goes by so quickly that’s another thing. Plus you get away with so many things because of it, especially when the bump starts showing which is a plus. 😊😊😊

Hi all,

I'm 35F, been ttc for 8 years. Diagnosed with PCOS 4years ago. I just started the LOCI trial treatment, my scan is on Friday. I am hopeful.
We just have to keep trying, I know it’s mentally and emotionally draining but it will all end in joy for us all.
I will keep you all updated.

Hi ladies,

Thanks for starting this thread. I am starting on the LOCI trial next week and have been in for my scan + blood tests and officially picking up my prescription tomorrow. I have been TTC for a year and recently found out I have "lean PCOS" with irregular cycles. I am hopeful that this trial will help me get my cycles more regular and hopefully get me pregnant.. and I also hope to be able to help more women by being part of this study.

Hope to share the journey with you all! How have you all been finding it so far? Sending babydust to all of you (and a little bit to myself too..!)

@Cherryblooms Welcome, I also have the “lean PCOS” not that I had ever heard of it before my diagnosis. Some side effects have been with me each month but, we keep trying!

I have just completed my 2nd month of medication after the first cycle where I didn’t ovulate and have ovulated this month!

My progesterone was 104 on the day they took it day 20/21. Glad to know I have ovulated but feeling so so frustrated.

I’m currently two days late for my period…but every test is negative and I’ve had period pain so I’m just waiting for it to arrive but it’s so mentally frustrating. To top this off I just wonder WHY won’t it happen if I’ve ovulated 😞

To add to this I have a Hycosy in two weeks so have been told to stop taking the medication and trying this month. This is even more frustrating as when I didn’t ovulate it took 8 weeks for me to have a period/ induced bleed to be able to start again.

sorey for the mixed update. Just seems like such a roller coaster and I hate not understanding my body.
