Culzean Castle and New Lanark could be fitted in en route to/from Largs/Glasgow.

Millport for the day on the ferry from Largs? Or Dunoon from Greenock?

I'd stay in Glasgow and go to Edinburgh for the day. Get the train or use one of the park & rides on the ring road. Edinburgh will be full really busy, especially the week of the English bank holiday. Do as FannyFifer says - any show that's been getting good reviews you'll have to book or will already be sold out. Loads of free stuff to look out for though.
