So, apparently I'm waiting for a hormone for night time training. My DS is 4.5 and still has a sodden nappy in the morning. When will I know he had the hormone and what is the hormone?

The hormone is an anti- diuretic called vasopressin. It reduces urine production at night.

You will know they are ready when morning nappies are either dry or it is clear that the wee is very fresh (i.e. warm) because it happened after they woke up. It can take until they are for 8 for the body to start producing it.
