Only aneuploid embryos after two rounds

loulamay · 28/02/2024 15:02

Looking for anyone who had only aneuploid embryos after PGT testing after two rounds but went on to have an euploid embryo in round 3!

first round, 16 eggs, 6 quality blasts - all aneuploid
second round, 6 eggs, 2 quality blasts - both aneuploid

I'm 43, so I realise age is against me but our clinic was very surprised and pleased at the number of eggs and very good-looking embryos from round one which is why we went again. Zero issues with husband's sperm.

We have a 2.5 year old daughter from previous IVF and I would so love another. We can afford another round which is what I think we're planning but Christ it's a slog.

Thank you for reading.

Anybody?

@loulamay hi, I am in a similar situation 🙈
Soon 42, ttc number 3 (second with my DH). Had 5 chemical pregnancies last 3 years, 2 EC, which gave 4 ubnormal (1st) and 2 ubnormal in the 2nd. All my hormones very good, very hood egg count (collected last time 17 and 10 fertilised), but no egg quality left
I am in the middle of third cycle, EC next Wednesday..
I do it for closure more than really think it will be different. I had 3 second opinion consultations, all said max one more and stop with OE. I just felt I needed last one as we planned 3, though chances are extremely low. I am doing DE route and adoption research and actually focusing more on those.
If we get any embryos we will test, so results will take a while (4weeks).
Sorry to hear you have similar struggles, it is really tough especially when you don't feel old and quantity is good....

loulamay · 01/03/2024 18:23

Thank you so much for replying @TT82 . We always planned 3 rounds as well. I have a friend in my village whose SIL is pregnant naturally at 43 and another friend from Cape Town who got pregnant with a healthy boy after an IUI at 44! So I feel like even if there's a small chance, I'll take it.

I have also started thinking about DE. How are you feeling about it? I am feeling pretty mixed at the moment.

TT82 · 01/03/2024 18:31

@loulamay oh I have a rollercoaster with DE... I am fine from just my egoistic side, mainly because I have 2 kids with my genetics. But when I start thinking about kid side, what that means, our family reaction....then comes anxiety.
But I needed to grieve my failures with previous 2 rounds and actually now I am in a much better state. I am a person that fies a thorough research unfortunately! So joined, listened and read sooooo many podcasts, views, information...
At least I realised that I am comfortable only with open ID, so UK or Portugal only (best for child). I am not in the UK, so would need to travel. But all this research and planning helps me not to think of OE failure 😞

loulamay · 01/03/2024 18:38

I also love to research! I'm a 'knowledge is power' type of gal.

We are currently with a Czech clinic and they only do anonymous donors, which I did wonder about. The main concerns are related solely to how the child would feel rather than how WE would feel. I have zero concerns that she or he would be loved, feel like our child, etc etc but what about the future and the DNA tests that you can do, and finding other half brothers and sisters....?!

Everything I've read so far (perhaps not as extensively as you, so if there are any faves like studies or links please feel free to send them on!) tells the same kind of story: as long as you're open from an early age and honest about it, using the right kind of language, then donor-conceived kids turn out JUST fine.

I actually think it will be something that's so common in the future that any stigmas around it will lessen massively.

TT82 · 01/03/2024 19:02

@loulamay I wish in Czech Republic they did open ID! We had a few discussions with the clinic there, really liked the dr, communication, everything.. (due to travel we stay with the clinic where we live).
But after listening to all these podcasts (Three makes baby; You look like me; Building a family)... I really don't feel comfortable with anonymous. In reality in future high chance they still will trace with dna websites... But after I listened of much struggle those people have to go through...yeah I think more of open ID (non-anonymous at 18)...
Once we close OE journey, I will arrange a few consignment to find out more details. I might change my view if in the UK long waiting time or no matching donor...or in Portugal similar

loulamay · 01/03/2024 19:19

Will have a listen of those podcasts. Thank you so much. Good luck with your retrieval - let me know how you get on!

Good luck for EC today! We spoke to our clinic and we will start our third and final round at the end of the month. Have been listening to Three makes baby and it's been so interesting. Thank you for the tip!

@loulamay hi, thank you! Just back, collected 22, but many were small so probably not mature. Last time I had 17 collected but only 2 embryos. We won't have any updates till mid next week when they tell us if any were biopsied for testing.. so now a waiting game.
Glad you found it useful! And good you are planning a third

loulamay · 06/03/2024 13:48

That's a great number though! Fingers crossed for you! The waiting is the worst x

How did you get on?

We're going again at the end of the month but I'm also doing more research into DE. There seems to be conflicting advice around the anonymous and open ID routes and I'm starting to feel confused about it all! Looks like the only European clinics that offer open ID are the UK and Portugal. I doubt we'll be able to afford UK prices. Ferticentro is a Portuguese clinic that seems to have good reviews. But then I was reading another post on here by a donor-egg-conceived child and she said the anonymity actually made life simpler because she couldn't contact the donor even if she wanted to, so there was no decision to be made.

@loulamay hi, we got update yesterday: 14 mature out of 22, 12 fertilised and we got 4 good quality biopsied: 4AA, 4AB, 6BB and 6day 4BB. I mean not amazing but so far best grading overall we have. We will know results in 4-5 weeks only, takes ages here.. Now I started thinking o we are improving overall, maybe still one more.. 🤦🏻‍♀️ though promised myself last EC
Regarding DE clinics, I fully understand you as conflicted myself. Ferticentro number 1 on ny list. Regarding open/anonymous...yeah we might not want them to contact their donor, but actually kids do want some, and then if they know there's this option, they might not rush or use. Otherwise they will agonise on these dna websites.. But agree that a question how easy to go with Portugal clinic etc. As yes UK will be very expensive.
If results are not good, we will book consultation with Ferticentro to find out more. I listened to their embryologist telling what information they provide about donors, time lapse incubators etc, and really liked..
Good luck with the next EC!! I will update in April where we are xx

TT82 · 04/04/2024 21:35

@loulamay just received our testing results, all uneuploid 😞 fully in pieces.. 3 rounds, 10 tested and nothing to transfer
Hope you get better news xx

loulamay · 06/04/2024 12:19

@TT82 Oh I'm so so sorry. Totally understand how you much be feeling. I've got my egg collection tomorrow and very apprehensive although scans are showing a good number of follicles so that's positive. But we had six embryos tested on the first round and not one single one came back as normal. The money down the drain! It's all so hard and so unfair. x

OP posts:
TT82 · 06/04/2024 19:44

@loulamay good luck tomorrow!! Stay positive, it is all luck which batch you get. So my negative experience doesn't mean anything for you . Lets hope for at least 1 good euploid one xx

loulamay · 07/04/2024 10:24

It's totally all luck! 15 eggs collected and all mature which is a good start. Now we wait!

OP posts:
TT82 · 07/04/2024 14:14

@loulamay good start!! 🙏 waiting the worst 😞

loulamay · 12/04/2024 12:10

It was going so well. On day 3 we had 9 embryos in the running still.

Had a day 5 report today - we have one hatching blastocyst that has been frozen ahead of testing but the others are 1xB (blastocyst), 3xEB (early blastocyst) and 2 compact Morula. I hadn't realised this but apparently, they can only do PGT testing on the hatching ones. Now feeling totally downhearted because if the B and EB don't catch up by tomorrow we only have one we can test. They will freeze those but we have no way to know if they're euploid.

I FRICKING HATE THIS. I also hate that I stupidly got my hopes up again and now it all feels like a potential failure.

OP posts:
TT82 · 12/04/2024 12:15

@loulamay oh just sending hugs 🥺 Fingers crossed they will catch up, some are hust slower sometimes
What do you feel about transferring not tested? I know high chance/risk to miscarry...but then there us a chance they can actually develop into a good pregnancies. I was listening to a good podcast NY clinic specialising in older over 40s women... They actually not for testing.. I was actually so upset I tested all and didn't try to transfer..

loulamay · 12/04/2024 12:43

I am terrified by the thought to be honest. I have had 4 miscarriages in total, and one D&C which showed an abnormality with the embryo. So to go through another miscarriage would be very hard.

My bigger worry though, is if the pregnancy developed, but we didn't find out until week 10 or onwards from NIPT testing, that there was something wrong and then we'd be faced with some difficult decisions.

I have also heard success stories and even mosaic embryos that self-correct after implantation.

I guess the good thing is that they won't discard them so we can leave the door open for now while we test the one HB that we have, and hopefully one or two more tomorrow if they make it.

How are you feeling about things now? This whole process is just so awful. Have you decided on another round?

TT82 · 12/04/2024 13:11

@loulamay we haven't properly sit down with ky husband to discuss all. I have called clinics in Greece, Portugal, but all say with history of no good quality it is a waste of time and money but if I wish they could try to change the protocol a bit etc. But all 3 pushed to DE..
It is just hard when numbers good.. there's this nudging feeling maybe next one.

I am also in a discussion with egg bank that has my heritage donnors, they could do open id (it will be bank responsibility not a government..so less reliable I guess, but I like a few matches).
We have a call wity my clinic here (we still haven't had a proper appointment) to discuss results and if anything can be done. But at this moment I am not sure I am mentally ready to continue trying with OE.

I really hope one of your slow growers will positively surprise you! Xx
