PGD failed to amplify ADN in 2 embryos

Hi.. I'm kind of hopeless now and can't find any information online. I was supposed to have a transfer today, it is a 5th day. On Saturday (3rd day) I was called from the clinic (name edited out by MNHQ) and informed that out of 6 embryons they managed to analyze (pgd) 5. One was okish, 2 were good and 2 had the best possible evaluation, according to the doctor. Today they called me to say that 2 were not viable, 1 was inconclusive and 2 were not analyzed due to the impossibility to amplify DNA.
I have searched and searched and can't find any information on this "failure to amplify DNA" and what the cause could be. The doctor told me that it could be either technical or biological.
They have analyzed all 3 of them again and were supposed to freeze at least 2.
Anyone with similar experience?

Hi OP so are these embryos from a previous egg collection? It's a bit confusing as PGD is usually done On day 5 blasts and the genetic results can take 2-6 weeks to get back so you have to do a freeze all cycle and transfer a couple of months later? Is this what you are doing?

I think when they say it was impossible to amplify dna it means that the embryo was too poor a quality to be able to extract the necessary cells?

PGD isn't normally done on day 3 embryos as the results are less accurate.....

It is my first cycle, had the egg extraction on last Wednesday, on Saturday (3rd day) they did the biopsy of the embryos to do a PGD testing (an express one, with results on Monday - yesterday). I was supposed to have a transfer yesterday, but since they couldn't amplify the DNA of these 2 embryos, they did the biopsy again and froze them and now they will have the results in 10 days and if either of those 2 is OK, they will do a FET.
I just found a whole procedure to be an emotional roller-coaster since I went from having the good news on Saturday ("2 embryos with the best possible evaluation") to "Failure to amplify DNA" on Monday.. and since I couldn't find any information on the internet on what the cause for this failure to amplify could be, thought to ask here.. Thank you so much for your input, if PGD is usually done on the 5th day, and they did it on the 3rd day, I guess I can still have a bit of hope that the problem is they did it too soon and not the viability of the embryos.

Wow never heard of them rushing through PGD to be able to do a fresh transfer!

The problem is that they have to basically do assisted hatching to do the biopsy and that's normally only possible if it's got to blastocyst stage (or day 5)

At day 3 the embryo should be around 8 cells so not much for the embryologist to biopsy from. At day 5 it's more like 100

Unless they are doing the new form of PGD which basically just takes a sample of the outer fluid rather than the cells which make up the placenta?

Perhaps the initial feedback you were given was actually the day 3 grading of the embryo rather than PGD result??

Or it sounds like they did the PGD on day 3 and then let them continue growing to day 5 and the quality has deteriorated over the 2 days......which is also the reason why PGD shouldn't be done at day 3 as no point testing embryos which would never have made blastocyst stage!

If I were you I'd be asking why they've gone down this route??

Just spoke to the doctor, she said this is how they usually do it if the reserve is good (in my case, the reserve/count was good, but the AMH extremely low (0.60), I am 41. My response to the stimulation was OK I guess (13 eggs, 6 embryos on the first day), but now it all depends on these 2 embryos that made it to the 5th day but didn't amplify on the 3rd day PGD. Well, I can only hope these 2 frozen embryos survive and are healthy, but it seemed today that she didn't really want me to have hopes. Oh well, not much I can do... Thankfully, I have a DS already, this was more like the last intent to give him a sibling...after waiting for 2 years after my cesarean, I then had a silent MC 3 years ago.

Hi, thanks for your interest! They did the 5 day biopsy of the 2 embryos, and froze them both. One isn't good, and the other one is genetically alright and morphologically 5AA. So I finally have 1 good embryo frozen! They'll do a FET on my first natural cycle. I really hope it works, though I'm afraid to hope..

I mean genetically normal

Hi, just wanted to update in case anyone finds themselves in a similar situation. Only one of my 2 frozen embryos was genetically ok and they transfered it mid February. I'm 9 weeks 5 days today! Had some brown spotting at 8 weeks, very little though, they put me back on Utrogestan again till 10 weeks. For now all seem to be going well. Will do Harmony in 2 weeks and if everything goes well hope to be able to sigh with relief finally. It already feels like a miracle.. The only thing is I have some unbearable nausea.

Just to update: I had my baby girl mid October by c-section! She's a healthy baby, big sleeper and feeds well! She is my little miracle, and all those people who told me "you only need one (embryo) for it to work" were so right! I hope this gives hope to someone out there and helps someone decide to give ivf a go - it was an emotional roller coaster but it was so worth it!
