When you wake up after a long hold.

I would say when you wake up in the afternoon. When the urine is most concentrated.

Just when you wake after your long sleep. It's about having held your urine for hours and not drank anything to dilute it

Do a 4+ hour hold and should be ok. FMU would be waking after sleep, though (if for baseline ovulation test) or same time each day.
