want to use a standard dk wool instead of cotton dk which is stated in teh pattern. How will this affect pattern, needles used and also will it be more or less wool used than cotton dk? Anyone help please?

You need to knit a test swatch - that is the only way that you will find out for sure what the knitted fabric looks like and what size needles you will need.

Yes, you need to do a tension square to check your gauge but there's no real reason why you can't substitue wool for cotton, although obviously the look will be different. Assuming your gauge is the same then you'll probably find that you need slightly less wool by weight.
