bespin, I'm afraid you're talking to a person who believes in absolutes. I do indeed accept that there are women who hold the philosophical belief that transwomen are women and therefore define themselves as lesbians if they sleep with a transperson with a penis.

They are wrong. The protected characteristic and therefore the legal definition of 'lesbian' is a woman attracted exclusively to her own sex. Transwomen are not of the female sex. Therefore they are not lesbians, however much pleading, coercing, wheedling or wordplay happens.

A trans woman might identify with women, that doesn’t mean they are a woman. I have had colleagues who have patients arrive in distress swearing until they are blue in the face that they are one sex when they are the other, until it is explained to them their sex determines their treatment and could be life or death. Why do my colleagues not get to decide (what is plainly obvious) in that situation? Why do students working in the team report them for that exchange?

Oh, right. You're asking if I would challenge a woman who claimed to be a lesbian but had a male partner? That would rather depend on the context. If she confided in me that she was married to a man but was closeted, then obviously, no, I wouldn't because her circumstances clearly would be socially complicated.
But an ostensibly out woman in a relationship with a man? Yep, the eyebrow would be arched and I'd probably have a conversation about it. Because there is nothing wrong with being Bi. It's not a dirty word. There is something wrong with pretending not to be Bi for some strange inclusive kudos reason.

Anyway, I've stayed up way past my bedtime and I really do have to get up at six, do a 9 hour shift and then come home and do all the shitwork in the home until I collapse in to bed again, and hope that my bastard period doesn't make a surprise appearance because I've run out of the big thick pads.
