Myrcella is poisoned but not necessarily dead - poisoned lipstick won't be as effective as the poisoned blades that the Sand Snakes use.

I suspect that Myrcella is very very dead. And Cersei is going to be majorly pissed at Jaime.

I know everyone says this when I favourite character goes but I really loved Jon, he was my main focus on the show, waiting for his scenes so I am not really bothered now. Will give it a go when Season 6 comes around but I won't be as into it now, which is for me because it was the best thing on TV. I read an interview with KH and it sounds pretty definite.
