Professor Brosens / sitagliptin

Is there anyone on here that's been treated with sitagliptin? Also anyone had the digital endometrium function test with professor brosens? I have an initial consultation Monday and then my biopsy next Friday

Does anyone know about long term effects of sitagliptin? Specifically, if we wanted to go for another child would we be likely to need it again or has it 'fixed' the problem for good?

@Laurenm1990sorry me again🤣🫣 did you call up before your consultation to book your appointment for the biopsy or they just had space to get you in quickly? And is it still 6 weeks for results? Trying to plan around work/travelling to Coventry and our consultation is around a week after I think I’ll ovulate, so just trying to work out the timings of everything and whether I should phone ahead to see if they’ll even have a slot xx

No need to apologise @ByDeftOpalZebra. I called them before I had my consultation about a week before and said I am due to ovulate 3 days after my consultation so wondered if I could book my biopsy in and they said yes and booked me in which was fab. My results took 3 weeks and 3 days to come in and they sent me the follow up consultation appointment. Much quicker than I imagined which I was grateful for. Always happy to answer any questions

Hi all, I wanted to share my success story with Sitigliptin in the hope it helps somehow.
First child born 2020, no issues conceiving
5 mc between 6-8 weeks trying for second, diagnosed hashimotos after the third and started levothyroxine. Polycystic left ovary but no PCOS.
Went for implantation clinic tests January 2024, biopsy showed bottom 5% on DEFT, started Sitigliptin for 3 months and break from ttc.
Conceived again the next cycle after finishing the course, also on progesterone protocol.
We did have some growth issues, whilst born healthy and now a chunky 1 year old she was growth restricted and placenta was visually in poor condition, no idea if this is related in any way.
Good luck to all, I really wish that you get your success stories too and that our journey provides some hope. It is a long, lonely road but all we can do is keep going x

@ByDeftOpalZebra hope your appointment went well and there's a plan for you! Did you by any chance ask if the sitagliptin effect is ongoing? Debating how and when i tell DH that i want number 3 😂

Hello,
I wanted to write on here because i have recently finished a 12 week course of Sitagliptin and after a second biopsy showed no progress on the deft. I’m gutted and feeling so so hopeless now. We’ve been trying for 2 years, multiple miscarriages. Brosens has recommended another 12 week course of Sitagliptin but I’m struggling to understand why this would have any positive effect.
Has anyone else had this where a second course helped? Has anyone had a third biopsy?

Yes - I saw very limited improvement of 1-5%. Took sitagliptin on and off for years - was scheduled to have a third biopsy in October - but am currently pregnant so haven’t had that. We ended up doing a deeper dive and looking into reproductive immunology with Dr Thum at the Lister Clinic. We found out that although I don’t have many NK cells - that they are very overactive, and am on a new protocol (intralipids, steroid) with Dr Thum. We’ve also been seeing Rachel Sherriff and she has been wonderful. So supportive, clear directions and hope- we’ve made subtle changes to diet, supplements and testing and treating microbiome. We have hope that this pregnancy will finally work out after 5 years and 8 losses since my daughter. Hope this helps, and feel free to ask any questions. The worst girl gang ever is a really helpful resource!

Thank you for your reply @Ezzyar and I really hope this works out for you, it sounds like you’ve been through so much. We’ve stopped trying for a year now because I can’t face more losses. Brosens has a new plan for me and I’m starting to feel a bit like an experiment so I’m going to also go back to my fertility clinic to see what else they can do. What other tests did you have? Keep us updated with how you get on! X

I am due to have my biopsy next week and I am very nervous. I am also going to the appointment alone which isn’t helping my nerves. I don’t do well with smears or embryo transfers as I struggle with the speculum. I usually do embryo transfers whilst sedated as I find it too difficult.

I know they recommend paracetamol before the appointment and they have gas and air available. If anyone has had the procedure, can you tell me what to expect? Do they use a speculum? Is it a similar length of time as a smear/ embryo transfer?

I went alone too, it was a while ago but what I remember is there being a very lovely assistant who kept me distracted. I found it better than a usual smear. The room had lower lighting than a GP and the whole experience was definitely more dignified. Best of luck to you x

Hello, they do use a speculum. There is a nurse and a doctor, and they are very kind and supportive. Tell them your worries and they will really talk you through how to rip through the gas and air. I found I didn't really even feel like I was in the room - which I found helpful (having had a breakdown whilst having a smear previously). Hope it goes as okay as it can. Deep breaths on that gas and air!

hey - sorry didn’t meant to leave you hanging! I’m now 9 weeks - and saw baby with heartbeat at 7 weeks. Things seem to be progressing well and I have another scan on Monday (9+3). Our losses kept on happening at 6/8 weeks or were chemicals - all to do with my autoimmune response. The tests that Prof Thum did looked in even more detail than Brosens at my immune / cellular response to pregnancy material. I finally feel less like an experiment and more like I have a solid bespoke medical protocol on my side. Even getting to this stage has proven that it has made a difference. And if this does result in another loss (although all symptoms are pointing to it not being a loss!) my protocol will be readjusted. I’m so much more positive than I’ve been for years. If anyone thinks there losses could be a result of autoimmune issues I’d really recommend speaking to Rachel Sherriff to help point you in the right direction. She does free 15 min phone calls. She’s changed our life. I’m also now in the process of getting an autoimmune diagnosis for suspected sjogrens - something that nobody else has picked up in over years, but that she highlighted in our first full session together.

Thank you so much for your responses @Happydays2025 and @Ezzyar! I had my biopsy today and boy I found it tough. But you were right - the nurse and Dr were lovely. The gas and air definitely helped to get me through it!
