Is there anyone on here that's been treated with sitagliptin? Also anyone had the digital endometrium function test with professor brosens? I have an initial consultation Monday and then my biopsy next Friday

Hi all I had my consultation today - he was lovely and took a lot of time to explain everything. I am due to have my biopsy next week. Hope everyone is getting on okay. I have heard that the results of the scratch only last for 3 months but obviously hoping something can be found in the biopsy as I have had 4 losses and the last one with progesterone clexane and aspirin x

Hi @Ttcafterlosses i had my biopsy a couple weeks ago and have an appt in 2 weeks to discuss the results, also had 4 losses and last one on progesterone.
I would recommend taking paracetamol before the biopsy as I found it quite uncomfortable, hope you get some answers! I will update this board when I have had my consultation for the results x

Hi wishingonastar90. Sorry for your losses. Thank you for the advice. Professor Brosens will be away so I will have the biopsy done by a colleague. Have you been advised not to TTC until you get the results? I know someone who had the scratch who mentioned the effects only last 3 months so waiting for the results obviously rats into that time

How do you mean it only lasts 3 months sorry? I haven’t been told not to ttc and have tried this month, my appointment will be the same time as I’m due on my period so will see what happens from there! X

Fingers crossed for your rainbow!

I know someone who had this procedure done after 6 losses and had a successful pregnancy straight after it. She mentioned that the effects of the scratch only last 3 months but this isn’t something mentioned by the consultants.

according to a quick google search on an nhs Liverpool hospital website it states
“In order for you to have an endometrial scratch, you will need to be seen by a clinician. It is normally performed between days 20 and 25 of your menstrual cycle and its effect can last up to 3 months.”

After my scratch I will have missed the fertile window so I am planning to try after the period following my scratch. I feel a little pressured to try to conceive in the following 3 months if this is the case.

Hi all I had the scratch/biopsy done this afternoon and my spotting is heavier than expected and having cramps. Did anyone else have the same?

Hey, yes I had some spotting and cramps but settled down over the next couple days, period came earlier also x

Hey all, I just wanted to give an update.. this cycle was the first cycle we were able to try following a 12 week course of Sitagliptin to treat low dEFT and low NK cells and I found out yesterday that I’m pregnant. Now given our history we’ll be taking it one step at a time, day by day but wanted to share to give some hope xxx wishing you all the best of luck, the biopsies are not nice but hopefully will really help us all going forward x

First of all, massive congratulations @Haz1990. Absolutely lovely news! It’s such a hard place to be but great to hear happy news

I wondered if anyone had any words of wisdom …

I was down in Coventry this week for biopsies (again, Prof Brosens was away but another doctor carried out the procedure). He was actually unable to take the biopsies though as he couldn’t gain enough access to my uterus due to a bend in it!?! I’ve also had this issue when having tests at my own clinic.

I have an online appointment with Brosens this week. He says he will do the test himself in another cycle but I’m not hopeful it will be possible. Does anyone know if any further pain relief is available at Coventry? I’m having Alice Emma etc tests done under sedation for this reason

I also owe an update. We've been through the sitagliptin treatment, I ended my 3 months in mid August. And we followed with a medicated FET which failed. We had a pause in September due to coming out of the medicated cycle. And then another medicated FET in October and were currently 4 weeks and 3 days pregnant, so we got our bfp. But time is going so slow and I'm desperate for our tests to show a dye stealer...maybe tomorrow.

It's our lowest quality embryo too but each one that survives the thaw is a chance.

Our official test date is tomorrow so I'll find out what happens next. But we're just taking it a day at a time as we've been here so many times before, albeit not through ivf.

I typically have a really thick lining and I think it's this which has caused our issues so I think it's the sitagliptin and the medicated cycle which has helped prepare my lining. As it was only 11mm at transfer on this round and previously its been 17, 18 and 22mm at different points of my cycle.

@Annabel82 have you spoken to the Prof about the Emma and Alice tests? He might save you a bit of money if you're not committed already. I'm not sure if they offer pain relief but it's worth phoning ahead to ask. His clinic are brilliant are really responsive to questions. I hope you're able to have the procedure so that you can get answers either way.

Congratulations on your result. We’ve just been through the process of embryo banking with pgta and it’s been interesting that our genetically normal embryos haven’t been the best looking/quality ones so fingers crossed

We’ve got another meeting with the Prof this week. He says he’ll do the biopsy on a future cycle but I’m not hopeful as two doctors have failed this week already. Apparently I have a sharp bend at the opening of my uterus which makes access with the instrument used very difficult whereas embryo transfer with a catheter is ok. I’m feeling very disappointed at the thought at not being able to have the test and desperately hoping there will be a way

If he's unable to carry out the biopsy you could ask if you can have the sitagliptin and take it empirically- it's a philosophy which seems to be heavily reliant on in infertility! It's worth an ask! And it's an old and stable medication that doesn't seem to clash with much. I Must add here, im not a medical professional...but we've all got our learnings from this journey haven't we!

Is the 82 bit of our name your birth year? If so we're around the same age too.

yep @MediaMinion, I’ll be 41 in Nov. My age has always been blamed although I feel like there must be other issues too. I’ve had two chemicals and one missed miscarriage. Tried naturally for two years ish. We did two failed ivfs and then resorted to embryo banking with pgta. Only 5 out of 16 embryos were euploid so I guess that backs up the age theory. They’ve been hard to come by so I’d like to make sure we’ve done implantation testing before transferring any.

That’s good advice about the meds- I’ll ask.

I hope you’re holding up ok. It’s a long and exhausting road, even with positive news x

@Annabel82 the dr also had difficulty getting to my liking and had to straighten my cervix out to get to it so it took longer than usual and was quite uncomfortable, they did offer gas and air before not during but I declined as I honestly wasn’t expecting it to be as painful as I found it, I hope prof Brosens is able to do the biopsy for you and would check with him about other pain relief that might be available, please keep us posted with how things go

my update; I had my results appt with prof Brosens today and my results were 71st percentile for progesterone (normal), 72nd percentile for DEFT test (normal) and 88th percentile for the nk cells test (normal) on report although verbally he said this was slightly high - so basically he hasn’t suggested a treatment plan as such for us which I’m not sure how I feel about, I’m relieved in one way but also confused in another as was sorting looking for an answer we haven’t found yet. I’m wondering if anyone would mind advising how this compared to their results please? Does this seem ‘normal’? I felt like they were all pretty high but I think the first 2 tests higher might be optimal? I’m really not sure I fully understand the percentiles!

My digital Deft was 17th percentile, indicating endometrium more akin to recurrent miscarriage failure rather than implantation failure, the latter is my issue.

I was expecting to have raised nk cells but no it’s actually what a lot of You have said about having an abnormal dEFT so going to pause TTC and try sitagliptin for 3 months.

I actually developed acute endometritis which resulted in sepsis very quickly after the biopsy/scratch which I was told has never ever happened since they started doing the procedure so that has delayed my journey but I will be starting Sitagliptin when I have been cleared by the clinic.

Hopefully we all get our babies soon xx

I'm so sorry to read what happened to you, but sounds as though you're gonna be fine. Fwiw, I had no known side effects from sitagliptin 🙂

hi all - sorry to hijack your thread. We have a video consultation with dr brosens after suffering our end max in a row a few weeks ago. Was just wondering if anyone who’s had one before could let me know what to expect - there wasn’t much info when we booked and want to make sure we don’t waste the time! @PADonk I really sorry I can't actually remember our first consultation. But if appropriate it will be followed with an appointment for a biopsy and then the results and a plan will follow that. The sitagliptin course is 3 months so you'll need to factor that into your plan if that's what's recommended for you.
