I am hoping for some advice.

I am currently working as a supply teacher full time in a y6 classroom. The behaviour is very challenging. Children screaming across classroom at one another, throwing pencils and bottles in anger, swearing, arguing with adults in the room as well as lack of respect. I have tried all sorts. Using TAs to take out certain individuals, moving seats, restorative conversations and following the school's behaviour chart. Prior to this, I was working as head of y7 so dealt with quite a lot of behaviour but I am really struggling and each day I come home drained. All staff in the school know how challenging this class is hence why it is open to supply. Any suggestions?

Leave. The fact that they are behaving so badly is an indication that the school is poorly managed. Difficult class or not, the behaviour you describe is indicative of a toxic school, where leaders have failed to step in and address such unacceptable behaviour. This tells me that things are unlikely to get any better. They have had at least four years to sort out the behaviour in that class. I would hazard a guess that there is bad behaviour across the rest of the school too. Your not under contract, I would make your excuses and get out of there personally.
