Toddler constantly trying to get to wfh DH

Toddler knows DH is there and constantly trying to get to him.

Is he in a separate room, does it have a lock on it?

Separate room, no lock.

It might be worth fitting a bolt or lock, in that case. Or using a stairgate to block access to the room.

It will be really rewarding to him if he sometimes succeeds in getting into the room, whereas if he always unsuccessful and you keep reiterating that Daddy will be back later, he is more likely to get bored and stop trying.
