What should I do now...?

First problem purchaser.

Listed auction item as collection only. Winning bid, lived at the other end of the country to me. I asked when she would like to collect it. Replied that they didn't realise it was collection only, would I post it. I agreed. This was all last Sunday (1st Dec).

Tuesday I sent her an invoice with a revised price including P&P, she still hasn't paid and it is Saturday now, nearly a full week since she won the item.

Do I have to escalate it as a non-payment? How do I do this, this is the first time I've ever had a problem on eBay!?

Thank you.

yep open a non payment dispute to get your fees back & close in 4 days then you can relist or do a second chance offer

You go to Ebay resolution centre & follow instructions there to file an unpaid item dispute. Be polite to your bidder at all times, they may still pay up.
