Can anyone advise on how this works. The person who won my item has not paid two days later and looking at their feedback, they have form for this. Can I do the second chance offer now or do I need to wait a certain time and report non paying buyer to ebayer first? Any advice would be great x

you really need to open a dispute and sort out the first buyer before offering it to the next person

yes open dispute for non pymt now, close 4 days later and only then do 2nd chance or relist - never before.
