However, the item is big (bike trailer) and this will involve considerable effort on my part (removing wheels and towing bar, plus 'wrapping'). He then wants to send a courier to collect it. He says he has bought 2 already this week already.

I really don't want the hassle to be honest. Do I have to sell it to him or can I withdraw from the sale as he didn't read collection only bit of my listing?

no - never, ever let a buyer arrange their own courier

You will have to cancel the sale to get your fees back, then you can relist

And only accept cash for collection too

Yep you have no obligation to do this and whilst he might be bona fide doing this is a classic step one of a scam (because if it goes through and he arranged the collection you will have no proof that it left your keeping in good condition, he claims it arrived damaged beyond repair and gets a refund and keeps the item). Cancel the sale.

Cancel. Even if its not a scam, you could have got higher bids if you had postage as an option. Someone did exactly the same to me; I relisted it with postage (being pedantic by blocking the original winner) and got twice as much for the item.
