What happens if an employee ( relative of the owner of the company) tests positive for covid but doesn't want to isolate and the owner is fine with that!?
The employees aren't happy but what can they do?

If they have tested positive, then they are breaking the law by not isolating.

I would assume that Test & Trace would be calling them daily to check they are at home, and to see how they are.

The issue is it's not anonymous so an employee could report and then when the employer discovers they've done that it's not worth the repercussions sadly.

They could contact either HSE or the council depending on which list the workplace lands on.

Look up your local authority public health team and report them. The type of employer that would do this will not be high paying and there are a lot of open jobs at the moment, it's an ideal time to get a new better job!
