I had a similar issue all through primary with my daughter requesting to go to the toilet all the time! Unfortunately whilst schools encourage "drinks on desks" they can then sometimes be quite strict about timing of allowing children to use the toilet and some children are slightly intimated to ask. My daughter did have a couple of infections and I would say trust your instinct and take your son for a check up as it is obviously concerning you and is different behaviour than normal. Also he is still young and they do get engrossed in play sometimes and forget, leaving it too long! My daughters paedatrician suggested she had anxiety and this made her bladder over active, has he been at school long? My daughter is in secondary now and has a toilet pass so that she can ask to leave the classroom at anytime no questions asked, its made her feel a lot better about the whole issue!

My son (also 4.5) has had a recurrent uti since NY day. He has been going to the toilet at similar intervals to your ds, but has been screaming in agony each time, and his urine has been extremely cloudy (almost muddy). Worth getting your ds checked, in case he has got a uti - you don't want him to get to the agony stage!

He would usually be a liitle sore when he wees if he does, worth getting checked anyway though.

Sorry but I have to disagree with the comments re him being sore when he has a wee! You shouldn't wait for this sign as not all uti's have that symptom, particularly if it is higher up the urinary tract or actually in the bladder or kidney.
