I think my conclusion is not to be offended as he had been drinking, just thinks it’s not very nice to say
Bit like saying, someone’s hair or outfit looks different

So he has to lie to her because it's her wedding day? Or would you prefer for him to say 'you know what I think, I dont like him'

I dont think it's that unusual to ask a bride if she is happy, especially once the ceremony and day celebrations are done and now she can relax a little.

Depending on how it was said I don't think I'd have taken it that way OP.

I'd have taken it as 'why are you even asking me if I'm happy on your Day? Today is all about you: I'm happy as long as you are happy.'

...or your male cousin is a bit of an arse.
