DS is in YR1. He has been left out frequently by one very popular boy since about last half term. I hoped a break over the summer would help to resolve things but it's still happening. It's also got worse as other DCs, when around this boy, are telling DS he can't play, to leave them alone. He plays apparently well with these other DCs when not with the one boy.

Today DS was told he couldn't play and in his own words other boy then threatened to slap him round the face if he kept asking. DS and other boy are normally friendly and I've had him round to play a few times and he is a very pleasant child.

So should I talk to the class teacher tomorrow or try to speak to their Mums first?

Please go to the school first.
I'm sorry your son is being treated like this.
As it's happening on school turf, I'm certain they would want to know.
Bullying is horrendous and they will want to deal with it.
Don't be fobbed off and don't let them downplay the bullying.

I feel so sorry for DS - even though he does have other DCs to play with. And want to slap the other boy myself (obviously won't though)!

Sorry you and your son are going through this. I speak from experience, please go through the school. As tempting as it may be do not discuss this with the parent, or other parents at the school. Good luck.
