Do mortgage lenders accept the income from a role if you are "acting up" into a position or is it your basic income from your substantive role that they count?

Tell them your actual income. Don't tell them you're "acting up". They don't need to know that.

You'll likely need payslips from 3-6 months so if you've bee acting up for less than that, you may need to explain the additional ££.

Also remember they want your income for a reason, to make sure you can afford it, so do be sure you can afford it once acting up finishes!

I honestly think that would be fraud. They'll presumably ask if you're temporary/on contract, so you would have to lie and say you're permanant in your role. Its really not worth it.
