Has anyone successfully retested embryos - PGS?

Hi all, we have two embryos frozen with our clinic. Unfortunately both of these embryos came back inconclusive - the <5% chance! One they couldn’t test at all as the tissue was so damaged so either sample has been damaged or it’s very abnormal. Other one came back suspected extra chromosome 22 but not to the extent they could declare it abnormal or mosaic.

im wondering if anyone has successfully retested and used the embryo - I’m worried even if one comes back normal that it then may not survive the defrost - biopsy - refreeze - defrost - transfer process!

I am afraid embryos won't survive the process.

This is my worry too so wondering if anyone has managed it!

@CocoPocoLoco I had an embryo retested (first biopsy came back as 'no result') and to my complete surprise it came back chromosomally normal. The embryologist told me that the embryo stayed the same grade (6BB - considered good quality at my clinic) and survived the thaw and second biopsy really well. There was apparenty no cell degradation and the embryo re-expanded and rehydrated really well too. I'm yet to have this embryo transferred and so I don't know how the embryo will do after a second thaw procedure and even then if it'll have what it takes to lead to a pregnancy and live birth. What I do know though is that thaw survival rates are very high these days, usually (95% and over). From what I've seen, second thaw survival rates are slightly lower, but not hugely so (around 93%).

A friend of mine re tested 3 previous "no results" and they all came back normal

CocoPocoLoco · 29/09/2022 20:56

@anotherdayanotherpathlesstravelled did any of them result in the pregnancy?

@CocoPocoLoco

I don't think they have transferred them yet as they used a family member as a surrogate with their final euploid but i know they plan to use them in the future hopefully for a sibling

I thought I would update this thread in case anyone else is looking for an answer. One of the embryos came back genetically normal and one mosaic but suitable for transfer. We transferred the genetically normal one. The embryologist said any impact of repeat biopsy and more freezing/thawing would be seen in the lab and it had defrosted and expanded well and started to grow again so she said no impact. I am currently nearly 11 weeks and have had a scan that showed a good size and strong heartbeat.

@CocoPocoLoco thank you for the update, I've just found this thread and it's giving me a bit of food for thought as we have one 'no result' embryo, I had kind of written it off in my head but it's nice to hear stories where it has worked out. Congratulations on your pregnancy and wishing you all the best

browneyedgirl626 · 07/08/2024 15:57

Also @CocoPocoLoco just wondering if inconclusive was the same as no result? Or are they two different things?

TumbleweedAgain · 08/08/2024 09:16

@CocoPocoLoco congratulations! And thanks for updating the thread. So great to read stories like this, it gives hope to others.

CocoPocoLoco · 09/08/2024 10:26

@browneyedgirl626 the PGS normal one was initially no result. The inconclusive did come back as mosaic. we were like you - initially wrote it off but then decided to retest as otherwise we would never use it so thought it was worth the risk re defrosting biopsying again etc.

@CocoPocoLoco that's great, yes I think it's worth the risk, I would probably retest over do an FET without knowing because I'm on intralipids so the whole thing becomes very pricey.

I'm actually in the process of doing an FET with my other euploid. One resulted in my baby boy and I'm hoping to transfer the other one this week. Still so nervewracking, I thought I would be less anxious second time around but I'm not lol

Terrapin22 · 10/08/2024 07:53

In case it helps, I had a “no result”, which I was told could mean that there was insufficient sample tissue. We decided to do a second biopsy. It came back genetically normal. I had it transferred and I’m over 5 weeks pregnant! To think that we were going to discard it because we thought it was an aneuploid… I suspect that a euploid will be strong enough to withstand the procedures. But the key is to ensure you have an excellent team of embryologists who are skilled at this.

CocoPocoLoco · 10/08/2024 08:28

@Terrapin22 I think something happened with our initial biopsies to have two out of 3 with no proper result - when chances of one with that are <5%. We were told that it could be bad sample or that it was so abnormal no result could be detected so was very surprised when it came back as euploid.

@browneyedgirl626 very best of luck with your FET. This is also for my second and agreed on the nerves. Took weeks to believe it had actually worked. Took us 5 years and 6 rounds for my first.
