Are Rosie and Suzie too similar to use for siblings?

Rose and Susan.

The names are not too similar, but the different spellings in the nicknames for the s/z sound looks a little odd. I think it would bother me, and possibly be confusing for them learning to spell their names.

They’re really similar in sound. The s and the z sound the same.
So many nice names out there! Do you already have a Rosie or a Suzie?

If you've got a Rosie, you could maybe have Susannah on the BC and shorten to Suzie? Or if you already have a Suzie, have Rose or another Rose name, like Rosanna, Rosalie, Rosa and nn Rosie?

But if you have (say) Rose and Susannah on the birth certs then they have options. Rosie snd Suzie are too nick namey for full names though nice enough as nns.

Which one do you have already? As nicknames for two sisters they are fine but they sound a bit matchy as given names imo. As pp mention Rose, Rosa or Rosalie with Susanna would work better.

Susie looks nicer written down, just the shape of the letters. And still makes the 'z' sound, same as Rosie.
