I currently work three days a week in a large secondary school. I'm a science teacher.

I've been considering increasing my hours but not sure I want to do that at the school I currently work in. Realistically would I get supply work for two days a week?

I'n in a similar situation to you. I work 3 days a week in a primary school and am available for supply teaching on Thursdays and Fridays. I get a mixture of pre-booked work and morning calls. I've been really busy on the run up to Xmas but generally I probably average 4 days supply a month. I really enjoy it and the extra money comes in handy!
