Have an application in for DD for September this year. Waiting to hear shortly. Since the application deadline a couple of circumstances have changed at our first choice school. It was a tough decision between first and second choice and now I am wondering if we have done the right thing.

If we are given our first choice school but are unhappy with this, what is the next stage? Do we go on a waiting list for second choice school if we ask?

Yes you can apply to go on the waiting list for the second choice school.

Yes, you can ask to go on the waiting list for your second choice school. I'm afraid that is your only realistic option.

Please do not reject the first preference school because of the changed circumstances, you need to keep your options open and the best way forward is to accept whatever is offered to you in the way of a school and then get on the waiting list and possibly appeal for a place at our second preference school.
If you reject the place then the LA as the admission authority will eventually find you another place but it could be miles away and it could be a school that you do not want your child to attend.

As admission says accept whatever school you are offered (1st, 2nd or 3rd choice). It is too late now to change the order of priority on your form.
If you get allocated your one you don't now want, go on waiting lists for the one/s you do want and hope a place becomes available.

The problems created by turning down a place can be very harsh (being left with no school at all or allocated one miles away).
Some people think turning down an offer might show the LA they are really serious about not wanting it and wanting another school instead. It doesn't work like that. The council has no discretion to bump you up any list - you join the list at a position that depends on how close you are to the school etc and get an offer if enough people turn down their place at that school (if they are moving away, going private or get a 'better' offer).

Good luck - hopefully you'll get your 2nd choice
